背景:小鼠间充质细胞在体外培养扩增困难,细胞凋亡是否是其原因之一,目前未知。目的:观察常氧和低氧条件下,小鼠肺间充质细胞的凋亡率是否有差别。方法:用组织块培养法获得C57BL/6J雄性小鼠肺间充质细胞,分别在常氧(体积分数20%氧)和低氧(体积分数5%的氧)条件下培养,观察细胞内活性氧水平、凋亡率和超微结构的变化。结果与结论:体积分数为5%和20%氧气培养的细胞随代数的增加造血细胞均逐渐减少。体积分数20%氧气培养的小鼠肺间充质细胞内的活性氧水平明显高于体积分数5%氧气培养的细胞。体积分数20%氧气培养的肺间充质细胞的凋亡现象较常见,体积分数5%氧气培养的细胞凋亡现象较少见。提示常氧培养条件下,细胞的凋亡率增加,降低氧浓度可减少细胞的凋亡率。
BACKGROUND: Mouse mesenchymal cells are difficult to culture and expand in vitro. Whether apoptosis is one of the reasons is unknown at present. OBJECTIVE: To observe whether there is a difference in the apoptosis rate of mouse lung mesenchymal cells between normoxia and hypoxia. Methods: Pulmonary mesenchymal cells of C57BL / 6J male mice were obtained by tissue culture method and cultured under normoxia (20% oxygen) and hypoxia (5% oxygen) respectively. The intracellular activity Oxygen level, apoptosis rate and ultrastructure changes. RESULTS AND CONCLUSION: The hematopoietic cells decreased gradually with the generation of algebra in 5% and 20% oxygen. The level of reactive oxygen species (ROS) in mouse lung mesenchymal cells incubated with 20% oxygen was significantly higher than that of 5% oxygenated cells. Apoptosis of lung mesenchymal cells cultured in 20% O2 is more common, and the phenomenon of apoptosis in 5% oxygen by volume fraction is rare. Prompt normoxic culture conditions, the cell apoptosis rate increased, reduce the oxygen concentration can reduce the cell apoptosis rate.
