为了提高木材类生物质的酶解率,提出了高温液态水两步预处理的方法。在底物浓度为5%,pH=4.8,50℃,40FPU/g底物的加酶量下,考察了不同预处理条件对桉木残渣酶解的影响,发现两步预处理后残渣的酶解率明显高于一步预处理后的酶解率,第二步预处理反应温度的升高和时间的延长都可提高残渣的酶解率,而温度的变化对其影响更明显。由于半纤维素和木质素的去除增大了纤维素与酶的接触机会,所以桉木经过180℃,20min和240℃,20min的两步高温液态水预处理后,72h的纤维素酶解率可达到97.20%,而只经过180℃,20min的一步预处理,72h的酶解率仅为72.81%。
