目的观察天花粉蛋白对人肺腺癌细胞株A549生长增殖的影响和在体外诱导A549肺腺癌细胞凋亡的效果。方法应用MTT法检测栝萎中提取的天花粉蛋白对A549细胞生长增殖的影响,DNA Ladder凝胶电泳及流式细胞术检测天花粉蛋白对A549细胞的诱导凋亡作用。结果经天花粉蛋白处理后A549细胞生长明显受抑,并呈一定的剂量时效依赖性;流式细胞仪直方图出现明显的亚二倍体峰,经TCS(40μg/ml)处理48h、72h后A549细胞的凋亡率分别为(27.00±2.08)%和(39.43±4.92)%,均高于顺铂组(2μg/ml)(P<0.01)。结论天花粉蛋白在体外能抑制A549肺腺癌细胞的生长繁殖,并能诱导该细胞发生凋亡。
Objective To observe the effect of trichosanthin on the proliferation of human lung adenocarcinoma cell line A549 and the effect of inducing apoptosis of A549 lung adenocarcinoma cells in vitro. Methods MTT assay was used to detect the effect of trichosanthin extracted from Fructus Lycii on the proliferation of A549 cells. DNA Ladder gel electrophoresis and flow cytometry were used to detect the apoptosis-inducing effect of Trichosanthin on A549 cells. Results After treatment with trichosanthin, the growth of A549 cells was significantly inhibited and showed a dose-dependent dose-dependent manner. The flow cytometry histogram showed a distinct hypodiploid peak. After treatment with TCS (40 μg/ml) for 48 h and 72 h, A549 cells were treated with A549. The apoptotic rates of the cells were (27.00±2.08)% and (39.43±4.92)%, respectively, which were higher than those of the cisplatin group (2 μg/ml) (P<0.01). Conclusion Trichosanthin can inhibit the growth and proliferation of A549 lung adenocarcinoma cells in vitro and induce apoptosis of the cells.
