目的通过对中药材茜草及其近缘种(易混品)的psb A-trn H基因序列进行分析,为中药材茜草的分子鉴定提供依据。方法提取中药材茜草及其近缘种的DNA,对叶绿体基因psb A-trn H序列扩增和测序,计算物种种内、种间Kimura 2-parameter(K2P)遗传距离。采用最近距离、相似性搜索、构建Neighbor-joining(NJ)系统聚类树3种方法进行鉴定分析。结果 psb A-trn H分子系统树支持中药材茜草不同居群样本聚为一单系分支,支持率为65%;中药材茜草与其近缘种psb A-trn H序列的种间遗传距离为0.00751~0.19338,远大于中药材茜草种内遗传距离0~0.00372。结论叶绿体基因psb A-trn H序列可以快速准确鉴别中药材茜草。
Objective To analyze the psb A-trn H gene sequence of Rubia cordifolia and its related species (mixed product), and provide evidence for the molecular identification of Rubia cordifolia. Methods The DNA of Rubia cordifolia and its related species was extracted and the psb A-trn H sequence of chloroplast was amplified and sequenced. The genetic distance of Kimura 2-parameter (K2P) within and between species was calculated. Neighbor-joining (NJ) phylogenetic tree was constructed based on nearest distance and similarity search. Results The psb A-trn H molecular phylogenetic tree supported different populations of Rubia cordifolia collected as a single branch with a support rate of 65%. The interspecific genetic distance between the Chinese traditional medicine Rubia cordifolia and its relatives psb A-trn H was 0.00751 ~ 0.19338, far greater than the genetic distance within the Chinese herbal medicine Rubiaceae 0 ~ 0.00372. Conclusion Chloroplast gene psb A-trn H sequence can quickly and accurately identify Rubia cordifolia.
