目的为了解高热对大鼠神经元的毒性作用,对高热预处理的温度———效应关系进行研究。方法①随机把50只大鼠分成5组,每组10只。A组:对照组;B组:40℃,15min(高热预处理);C组:41℃,15min(高热预处理);D组:42℃,15min(高热预处理);E组:建立全身热疗模型:动物全身加热使直肠温度达42℃(2h)。②WBH治疗:预处理24h后,戊巴比妥钠麻醉下行WBH治疗,24h后行神经功能损害评估,处死动物,制作脑片,HE染色,观察神经元损伤情况,以确定预处理的最佳温度。结果B、C、D、E组CGNs发生改变,以E组为甚;B、C、D组CGNs DNA琼脂糖凝胶电泳显示:DNA梯状条带呈递减。
Aim To understand the toxic effect of hyperthermia on neurons in rats, the temperature-effect relationship of hyperthermia preconditioning was studied. Methods ①50 rats were randomly divided into 5 groups, 10 in each group. Group A: control group; group B: 40 ℃, 15 min (hyperthermia pretreatment); group C: 41 ℃ for 15 min (hyperthermia pretreatment); group D: 42 ℃, 15 min Hyperthermia model: The whole body was heated so that the rectum temperature reached 42 ℃ (2h). WBH treatment: After 24h pretreatment, WBH treatment was performed under pentobarbital sodium anesthesia, neurological impairment was assessed 24h, animals were sacrificed, brain slices were made and HE staining was performed to observe the neuronal damage to determine the optimal temperature for pretreatment . Results CGNs changed in groups B, C, D and E, which was more than that in group E; CGNs DNA agarose gel electrophoresis in groups B, C and D showed that the DNA ladder was decreased.
