目的探讨半夏泻心汤对H_2O_2诱导的RIN-m5F细胞凋亡的影响及其机制。方法大鼠随机分组灌胃,取血收集含药血清,300μM H_2O_2干预RIN-m5F细胞,测定半夏泻心汤最佳浓度,后随机分组,流式细胞术检测各组细胞凋亡率、Western blot检测各组蛋白表达。结果半夏泻心汤含药血清最佳浓度为10%(P<0.05);与模型组比较,半夏泻心汤含药血清可显著降低RIN-m5F细胞凋亡率(P<0.05),下调Caspase-3、Bax的表达(P<0.05),上调Bcl-2、PAKT的表达(P<0.05)。结论半夏泻心汤含药血清可抑制RIN-m5F细胞凋亡,其机制可能与PI3K/AKT通路的活化有关。
Objective To investigate the effect and mechanism of Banxia Xiexin Decoction on H 2 O 2 -induced apoptosis of RIN-m5F cells. Methods The rats were randomly divided into three groups. The serum was collected from the rats and the serum was collected. 300μM H 2 O 2 intervention was performed on the RIN-m5F cells. The optimal concentration of Banxiaxiexin Decoction was determined. The apoptosis rate of each group was detected by flow cytometry blot detection of protein expression in each group. Results The optimal serum concentration of Banxiaxiexin Decoction was 10% (P <0.05). Compared with the model group, the serum of Banxiaxiexin Decoction could significantly reduce the apoptosis rate of RIN-m5F cells (P <0.05) Downregulate the expression of Caspase-3, Bax (P <0.05), up-regulate the expressions of Bcl-2 and PAKT (P <0.05). Conclusion The serum containing Banxiaxiexin Decoction can inhibit the apoptosis of RIN-m5F cells, which may be related to the activation of PI3K / AKT pathway.
