目的 探讨中期因子 (MK)mRNA在原发性肝细胞癌 (HCC)中的表达及其意义。方法 收集 33例人肝细胞癌组织、配对癌旁肝组织及 1 0例肝良性肿瘤组织、5例正常肝组织 ,采用TRIzol一步抽提法提取总RNA ,应用逆转录 聚合酶链反应 (RT PCR)检测MKmRNA的表达。结果 2 8例肝细胞癌可见MKmRNA表达 ,阳性率为 84.8% ,而配对的癌旁组织中仅见 3例表达 ,正常肝脏和肝脏良性肿瘤组织无 1例阳性。统计学分析表明 ,肝细胞癌MKmRNA表达率与其他各组的差异有非常显著性 (P <0 .0 1 ) ,但肝细胞癌MKmRNA表达与其组织学类型、病理分级、瘤体大小、包膜状况及其甲胎蛋白 (AFP)值之间差异无显著性 (P >0 .0 5)。结论 肝细胞癌在mR NA水平上表达MK增加 ,检测MK表达情况可作为HCC的辅助诊断指标之一 ,对术后制定进一步治疗方案也有参考价值
Objective To investigate the expression of midkine (MK) mRNA in primary hepatocellular carcinoma (HCC) and its significance. Methods Totally 33 human hepatocellular carcinoma tissues, matched paracancer liver tissues, 10 benign liver tumor tissues and 5 normal liver tissues were collected. The total RNA was extracted by TRIzol single-step extraction and analyzed by reverse transcriptase-polymerase chain reaction ) For the detection of MK mRNA expression. Results The expression of MK mRNA was found in 28 cases of hepatocellular carcinoma with a positive rate of 84.8%. Only 3 of the paired paracancerous tissues were expressed, and none of the normal liver and benign liver tumor tissues were positive. Statistical analysis showed that the expression of MK mRNA in hepatocellular carcinoma was significantly different from other groups (P <0.01), but the expression of MK mRNA in hepatocellular carcinoma was not related to the histological type, histological grade, tumor size, There was no significant difference between the status and AFP value (P> 0.05). Conclusion The expression of MK increased at mR NA level in hepatocellular carcinoma. Detecting the expression of MK may be one of the auxiliary diagnostic indexes for HCC, and it is also a reference value for further treatment plan after operation
