目的 探索人眼视神经星形胶质细胞体外培养方法 ,建立细胞系 ,为进一步研究视神经损伤后的修复提供充足的细胞来源。 方法 用组织块培养法、0 .2 5 %胰蛋白酶和 0 .0 6 % EDTA混合消化液取材培养法 ,对婴儿眼视神经星形胶质细胞进行培养 ,观察细胞形态和生长特性 ,并进行免疫组织化学鉴定。 结果 采用组织块培养的细胞 ,生长较缓慢。用混合消化液培养的细胞 ,贴壁早、生长快 ,约 7d细胞基本融合。培养的细胞胶质纤维酸性蛋白 (glial fibrillary acidic protein,GFAP、S- 10 0蛋白及 Vimentin染色阳性 ,CD34染色阴性。 结论 用混合消化液培养法可成功培养出人眼视神经星形胶质细胞。
Objective To explore the method of in vitro culture of human optic nerve astrocytes and to establish cell lines, which can provide sufficient source of cells for the further study of optic nerve injury repair. Methods Tissue explant culture, 0.52% trypsin and 0. 6% EDTA mixed digestion culture were used to culture the optic nerve astrocytes in infants. Morphology and growth characteristics were observed and immunized Histochemical identification. Results Tissue cultured cells grew more slowly. Cells cultured with mixed digestive juice, adherent early, fast growth, basic fusion about 7d. Glial fibrillary acidic protein (GFAP, S-100 protein and Vimentin positive staining, negative for CD34 staining) .Conclusion Human optic nerve astrocytes can be cultured successfully by mixed culture of digestive juice.
