我们在制备HSA药箱中,做了HSA及SnCl_2不同浓度的试验,其鉴定方法为纸层析上行法,展开剂用85%的甲醇,发现HSA浓度在5-50mg对标记率没有影响,标记率达95%以上;SnCl_2浓度在0.25—5mg,标记率均达95%以上。但pH值对标记率有明显影响,在pH>5时标记率明显下降,pH<4时标记率稳定,说明SnCl_2浓度及HSA浓度对此实验要求不很严格,而~(99m)Tc-HSA溶液的最终pH使它是直接影响标记率的主要因素。
In the preparation of HSA kit, HSA and SnCl 2 were tested in different concentrations. The identification method was paper chromatography with 85% methanol. It was found that the concentration of HSA in the range of 5-50 mg had no effect on the labeling rate. Rate of 95% or more; SnCl_2 concentration of 0.25-5mg, labeling rate reached more than 95%. However, the pH value had a significant effect on the labeling rate, markedly decreased at pH> 5 and stable at pH <4, indicating that the experimental conditions of SnCl_2 and HSA concentrations were not very stringent, while ~ (99m) Tc-HSA The final pH of the solution makes it a major factor that directly affects the labeling rate.
