目的:制备兔抗伤寒杆菌Fe-SOD血清,并研究其对鼠伤寒杆菌攻击小鼠的免疫保护作用,探讨SOD与沙门氏菌致病性的关系。方法:用纯化的伤寒杆菌Fe-SOD免疫家兔,制备兔抗SOD血清,并用ELISA法检测抗血清效价;免疫电泳、双向琼脂扩散试验和抗血清对酶活性抑制试验检测抗血清的特异性。将所制备的抗血清与鼠伤寒杆菌混合,放37℃30m in,经腹腔注入小鼠体内,并观察3 d和7 d 小鼠的存活情况,以确定抗血清的免疫保护作用。结果:抗血清的ELISA效价为1:10240,免疫电泳结果显示抗血清与纯化的伤寒杆菌Fe-SOD及无细胞溶解物在相同位置各只形成一条沉淀线;双向琼脂扩散试验结果显示抗血清不与牛红细胞SOD形成沉淀线,但抗血清对酶活性抑制试验结果显示抗血清可抑制24.92% 的红细胞SOD活性,说明伤寒杆菌Fe-SOD与牛红细胞SOD有低度交叉反应。抗血清可抑制42.58% ~73.38% 的沙门氏菌无细胞溶解物中SOD活性。经抗血清处理过的5LD50鼠伤寒杆菌攻击的小鼠3d 和7d 的存活率分别为90% 和60% ,均明显高于对照组(P< 0.01或P< 0.05)。结论:抗伤寒杆菌Fe-SOD血清对受鼠伤寒杆菌攻击的
OBJECTIVE: To prepare rabbit anti-Salmonella typhimurium Fe-SOD serum and study its immune protection against Salmonella typhimurium-challenged mice and to explore the relationship between SOD and Salmonella pathogenicity. Methods: Rabbit anti-SOD serum was prepared by immunizing rabbits with purified Salmonella typhi Fe-SOD and the antiserum titer was detected by ELISA. The specificity of antisera was tested by immunoelectrophoresis, two-dimensional agar diffusion test and antisera on enzyme activity inhibition test . The prepared antiserum was mixed with Salmonella typhimurium and put at 37 ℃ for 30 min. The mice were injected intraperitoneally into the mice and the survival of the mice was observed on the 3rd and 7th days to determine the antiserum protective effect. Results: The ELISA titer of antiserum was 1: 10240. The result of immunoelectrophoresis showed that the antiserum formed a precipitation line at the same position with purified Salmonella typhimurium Fe-SOD and cell-free lysate. The two-way agar diffusion test showed that antiserum The results showed that the antiserum could inhibit 24.92% SOD activity of erythrocytes, indicating that the Salmonella typhi Fe-SOD and bovine erythrocyte SOD low cross-reaction. Antiserum inhibited SOD activity in 42.58% -73.38% Salmonella cell-free lysate. The survival rates of mice challenged with 5LD50 Salmonella typhimurium treated with antiserum for 3d and 7d were 90% and 60%, respectively, which were significantly higher than that of the control group (P <0.01 or P <0.05).
