Spatial localization of S,Cl,K,Ca,Ti,Mn,Fe,Ni,Cu,Zn and Cd in frozen-hydrated sections of Sedum plumbizincicola,a Cd/Zn hyperaccumulator from a mine area of Chunan county,Zhejiang Province, east China, was quantitatively determined using cryo-micro-PIXE(Particle-Induced X-ray Emission). The shoot of S.plumbizincicola hyperaccumulates up to 1470 mg kg-1 of Cd and 14600 mg kg-1 of Zn(dry weight)in field-grown plants.Micro-PIXE analyses showed that in the root, the surface or/and epidermis accumulate high concentrations of Mn,Fe,Ti,Cu and Ca,and Zn is concentrated in the cortex but deplete in the central cylinder. In the stem,K and Ca show the opposite distribution pattern among various tissues.Zn is preferentially accumulated in the stem epidermis(5088 mg kg-1 frozen-hydrated weight)and also in the vascular bundles and neighbouring parenchyma cells of pith.Interestingly, some of the vascular bundles of stem are depleted in Zn.Stem Cd is preferentially accumulated in the epidermis(550 mg kg-1)and evenly distributed in the cortex and central cylinder(213 and 132 mg kg-1).Taking tissue area(or weight)into account,22.1%,46.2%and 31.8%of total Zn and 17.1%,71.6%and 11.3%of total Cd in the stem are distributed in epidermis, cortex and central cylinder, respectively. In the leaf,Zn is preferentially accumulated in the epidermis and Cd is homogeneously distributed among various tissues. It was calculated that 81.3%of total leaf Cd is distributed in palisade and spongy mesophyll. Therefore, the parenchyma cells,e.g.cortex in stem and mesophyll in leaf, are the major storage and detoxification sites for Cd and Zn in S.plumbizincicola.
