一、引言 NK细胞为“MHC非限制性”,“裸细胞”,“大颗粒淋巴细胞”.NK细胞的发育过程不甚明了.外周血及器官中的成熟NK功能主要根据对某些肿瘤细胞(如人的K562、小鼠YAC-1)的天然杀伤能力而定性定量.上述诸项NK的基本概念近几年均有惊人发展,其在抗癌免疫反应中可探索的空间均得以较大扩展.二、NK细胞的发育生物学1、NK和T、B细胞的共同前体细胞的发现 淋巴类细胞系决定着T、B和NK细胞的产生.人们一直尽力寻找这三类细胞的共同前体细胞.发现小鼠编码锌指纹蛋白(IKAROS)的基因发生突变或Knockout后,HSC向淋巴类细胞的分化被中止,其它造血细胞的分化不受影响,推测IKAROS基因的表达涉及HSC向淋巴类细胞的分化.2、NK细胞与T细胞分化发育的关系 联合重症免疫缺陷综合征(SCID)病人体内缺乏T细胞和NK细胞,但含有B细胞和其它髓样细胞,提示人类T细胞和NK细胞含有共同前体细胞;人类胚胎未成熟NK可表达CD3的所有四条链的基因.成熟NK受IL-2激活仍可表达CD3分子.NK细胞所承担的细胞毒效应与CTL有相近之处,NK细胞分泌细胞因子的种类及数量与Th细胞相近,故倾向认为NK细胞和T细胞为个体发育上关系十分接近的两类细胞.
