目的检测新培育的IRM-1近交系小鼠的基因纯合性,并与IRM-2、615近交系小鼠进行比较,确定其遗传特性。方法按照国家标准GB/T14927.1-2001、GB/T14927.2-2001遗传检测操作规程,对IRM-1近交系小鼠进行Akpl等25个遗传生化标记基因纯合度、组织相容性基因纯合度的测定,用同系异体间进行移植试验;以及毛色基因测试。结果IRM-1近交系小鼠的25个遗传生化标记基因均为纯合体,其中有5个生化标记基因多态性位点与IRM-2近交系小鼠不同,有8个生化标记基因多态性位点与615近交系小鼠不同;皮肤移植100d后,未见排斥现象;毛色基因测试,F1的毛色全部为野生色,基因型为aaBBCCDD。结论IRM-1小鼠遗传纯合度已达国家标准,是基因位点高度纯合的近交系小鼠。
Objective To detect the gene homozygosity of newly-cultured IRM-1 inbred mice and to compare their genetic characteristics with IRM-2,615 inbred mice. Methods According to the national standard GB / T14927.1-2001 and GB / T14927.2-2001 genetic testing procedures, 25 inbred strains of IRM-1 inbred mice were homozygous for histochemical markers such as Akpl, histocompatibility gene Homozygosity was measured using allogeneic transplantation test; and coat color gene test. Results All 25 genetic markers in IRM-1 inbred mice were homozygous. Among them, 5 biochemical marker gene polymorphisms were different from IRM-2 inbred mice and 8 biochemical marker genes Polymorphic loci were different from 615 inbred mice; no rejection was observed after skin transplantation for 100 days; for the coat color gene test, F1 coat color was all wild color and the genotype was aaBBCCDD. Conclusion The genetic homozygosity of IRM-1 mice has reached the national standard and is a highly homozygous inbred mouse with genetic loci.
