研究我国艾滋病病毒Ⅰ型 (HIV 1)病毒样颗粒 (VLP)候选疫苗的免疫原性 ,为疫苗进行灵长类实验提供实验依据。将 gag和gag -V3VLP不同剂量 (5 0 μg、10 μg、1μg)在有佐剂 (氢氧化铝 )和无佐剂条件下皮下免疫小鼠 ,然后眼眶采血 ,用ELISA法观察免疫鼠血清中抗体与剂量关系 ,以及中和抗体滴度和抗体IgG亚型IgG1和IgG2a的水平。同时取鼠脾淋巴细胞 ,体外抗原刺激后收取淋巴细胞分泌上清 ,检测细胞因子IFN γ和IL 5水平。结果是gagVLP免疫剂量与是否加佐剂对免疫效果没有显著性差异 (各组P >0 1) ,而 gag -V3VLP免疫鼠剂量与加入佐剂有显著性差异 (各组P <0 0 1)。gag和 gag -V3VLP(5 0 μg ,佐剂 )中和抗体滴度为 1∶2 0和 1∶40 ,同时gag和gag-V3VLP免疫鼠血清 (5 0 μg ,佐剂 )IgG2a和IgG1均有升高 ,IgG2a稍高于IgG1;细胞因子 (5 0 μg ,佐剂 )IFN γ(gag2 40 0 pg/ml,gag -V3 10 0 0 pg/ml)和IL 5 (gag 40 0 pg/ml,gag -V3 2 0 0pg/ml)均有升高 ,IFN γ升高高于IL 5。由此得出 :HIV 1gag和gag -V3VLP候选疫苗能刺激机体产生抗体 ,其抗体有中和活性 ;抗体亚型IgG2a稍高于IgG1,Th1和Th2细胞分泌的细胞因子 (5 0 μg ,佐剂 )IFN γ比对照组升高 16 0倍 ,IL 5比对照组升高 80倍。表明VLP疫苗能诱导机体?
To study the immunogenicity of candidate vaccine of HIV 1 virus-like particle (VLP) in China and to provide experimental evidence for primate vaccine. Mice were immunized subcutaneously with different doses of gag and gag-V3VLP (50 μg, 10 μg, 1 μg) in the adjuvant (aluminum hydroxide) and without adjuvant, and the orbital blood was collected. The serum was detected by ELISA Antibody to dose relationships, as well as neutralizing antibody titers and levels of IgG subclass IgG1 and IgG2a. At the same time, splenic lymphocytes were taken from the mice and the supernatant was collected from the lymphocytes stimulated by antigens in vitro. The levels of cytokines IFN-γ and IL-5 were detected. The results showed that there was no significant difference between the immunized doses of gagVLP and those of adjuvants (P> 0 1 in each group), while the doses of gag-VVVV immunized rats were significantly different from those of adjuvants (P <0.01). Neutralizing antibody titers of gag and gag -V3VLP (50 μg, adjuvant) were 1: 20 and 1:40, respectively, and both gag and gag-V3VLP immunized rat serum (50 μg, adjuvant) IgG2a and IgG1 IgG2a was slightly higher than that of IgG1; cytokine (50 μg, adjuvant) IFNγ (gag2 40 0 pg / ml, gag-V3 10 0 0 pg / ml) and IL 5 gag -V3 200 pg / ml) increased, IFN γ increased higher than IL 5. The results showed that: HIV 1gag and gag -V3VLP vaccine candidates can stimulate the body to produce antibodies, the antibodies have neutralizing activity; antibody subtype IgG2a slightly higher than IgG1, Th1 and Th2 cells secrete cytokines (50 μg adjuvant ) IFN γ increased 16 times higher than the control group, IL 5 increased 80 times than the control group.
