目的:建立高效液相色谱法同时检测蔬菜中赤霉素GA3、GA4和GA7的方法。方法:采用80%甲醇对样品进行提取,经乙酸乙酯萃取,C18固相萃取小柱净化,旋转蒸发仪浓缩,ZORBAX Eclipse XDB-C18反相色谱柱,以0.5 ml/min流速,乙腈/水(p H 3.0)梯度淋洗,二极管阵列检测器检测,外标法定量,保留时间及光谱图定性。结果:方法检出限GA3:0.016 mg/kg;GA4:0.013 mg/kg;GA7∶0.016 mg/kg,加标回收率为76.8%~92.3%,相对标准偏差在2.6%~7.6%之间。结论:方法准确可靠、快速,灵敏度较高,易于推广,适用于蔬菜中赤霉素GA3、GA4和GA7的检测。
Objective: To establish a method for simultaneous determination of gibberellin GA3, GA4 and GA7 in vegetables by high performance liquid chromatography. Methods: The samples were extracted with 80% methanol, extracted with ethyl acetate, cleaned up with C18 solid phase extraction cartridge and concentrated on a rotary evaporator. The residue was chromatographed on a ZORBAX Eclipse XDB-C18 reverse phase column with acetonitrile / water (p H 3.0) gradient elution, diode array detector detection, external standard quantification, retention time and spectral characterization. Results: The detection limits were GA3: 0.016 mg / kg, GA4: 0.013 mg / kg and GA7: 0.016 mg / kg, respectively. The recoveries were 76.8% -92.3% with relative standard deviations between 2.6% and 7.6%. Conclusion: The method is accurate, reliable, rapid, sensitive and easy to popularize. It is suitable for the determination of gibberellins GA3, GA4 and GA7 in vegetables.
