泛素样含PHD和环指域蛋白1(UHRF1)是UHRF家族的重要成员。生理情况下,UHRF1参与调节淋巴细胞的发育和功能,并在维持子细胞的甲基化状态、细胞生长以及维持基因组稳定等方面也发挥重要作用。UHRF1在多种肿瘤中呈高表达,且其表达与患者的临床病理特征及不良预后密切相关。UHRF1可通过表观遗传学机制对多种肿瘤抑制基因的表达调控而实现对肿瘤细胞的生长、侵袭转移、氧化应激及DNA损伤修复等特性的调节。鉴于UHRF1在肿瘤中的特异性高表达及对细胞生长的重要作用,UHRF1有望作为抗肿瘤治疗的有效靶点。
Ubiquitin-like PHD and ring finger domain protein 1 (UHRF1) are important members of the UHRF family. Physiologically, UHRF1 is involved in the regulation of lymphocyte development and function, and plays an important role in maintaining the methylation status of cells, cell growth and maintaining genomic stability. The expression of UHRF1 is highly expressed in a variety of tumors, and its expression is closely related to the clinicopathological features and poor prognosis of patients. UHRF1 can regulate the growth, invasion and metastasis, oxidative stress and DNA damage repair of tumor cells by regulating the expression of many tumor suppressor genes through epigenetic mechanism. Given the high expression of UHRF1 in tumors and its important role in cell growth, UHRF1 is expected to serve as an effective target for anti-tumor therapy.
