目的探讨大黄素对人胰腺癌细胞株Panc-1增殖和凋亡的影响。方法人胰腺癌细胞株Panc-1经不同浓度大黄素(10、20、40、80、160μmol/L)分别作用24、48、72 h后,应用CCK-8法检测细胞增殖,光学显微镜下观察细胞形态学改变,流式细胞术检测细胞凋亡。结果大黄素对Panc-1细胞的增殖抑制作用呈明显的浓度和时间依赖性,20、40、80μmol/L大黄素作用Panc-1细胞24 h后凋亡率分别为7.1%、15.2%、21.4%。结论大黄素可显著抑制人胰腺癌Panc-1细胞生长,大黄素对人胰腺癌Panc-1细胞株的增殖抑制作用可能以诱导细胞凋亡方式为主。
Objective To investigate the effects of emodin on the proliferation and apoptosis of human pancreatic cancer cell line Panc-1. Methods Human pancreatic cancer cell line Panc-1 was treated with different concentrations of emodin (10, 20, 40, 80 and 160μmol / L) for 24, 48 and 72 h respectively. Cell proliferation was detected by CCK- Morphological changes were observed by flow cytometry. Results Emodin inhibited the proliferation of Panc-1 cells in a time-and concentration-dependent manner. The apoptotic rates of Panc-1 cells treated with 20, 40 and 80 μmol / L emodin for 24 h were 7.1%, 15.2% and 21.4 %. Conclusion Emodin can significantly inhibit the growth of Panc-1 cells in human pancreatic cancer. The inhibitory effect of emodin on the proliferation of Panc-1 cells may be mainly induced by apoptosis.
